ABSTRACT The aim of this study was to evaluate the antifungal action of ethanolic extracts of avocado seeds, by conducting two experiments in the laboratory of plant pathology of EPAMIG SUL/Lavras, MG, Brazil, in a completely randomized design with three replicates. The purpose of this work was to reduce the cost and improve the quality of the plant material during the micropropagation process in tissue culture. Partially improvement in the rooting process, coupled with cost reduction was obtained during the invitro rooting by the use of a natural support based on sugarcane bagasse as a substitute for the traditionally used agar gelled medium. Alveolus-capillary permeability on both normal and HIV seroreactive individuals BABT

The aim of this study was to evaluate the alveolus-capillary permeability by the lung clearance rate of 99mTc-DTPA(Technetium99m-diethylene triamine penta-acetate), (LCR-DTPA), both in normal and in asymptomatic HIV seroreactive patients. Thirty individuals were studied, 21 seronegative normal volunteers and 9 HIV seroreactive patients presenting normal chest radiography and no respiratory infection symptoms. LCR-DTPA was determined by inhaling 99mTc-DTPA and obtaining images in a gamma camera. The 99mTc-DTPA clearance rate in normal individuals was 0.99±0.15%.min-1 and in patients 2.31±1.25%.min-1. Studies were performed to analyze the genetic characterization using RFLP-ITS and Intron (primer EI1) markers and the amplification of the cap20 pathogenicity gene by PCR in Colletotrichum gloeosporioides isolates of different hosts plant. The genetic variability was accessed using RFLP-ITS and Intron markers and grouping by UPGMA method. This work describes an adapted thermal-gradient block with internal illumination, allowing experiments with light-requiring seeds. The temperature dependence study on the germination of the photoblastic Plantago tomentosa seeds was carried out in order to test the performance of the apparatus. With the aid of the model described here, some parameters related to the response of the seeds to temperature could be estimated, showing that the apparatus was suitable for the studies on the temperature dependence of photoblastic seeds.

Four variants of the potassium acetate procedure for DNA extraction from ixodid ticks at different stage of their life cycles were evaluated and compared with phenol-chloroform and ammonium hydroxide methods. The most rapid and most efficient variant was validated in the DNA extraction procedure from the engorged ticks collected from bovine, canine as well as from house ticks for the screening of Borrelia burgdorferi sensu lato, Anaplasma spp. and Babesia spp. The ammonium hydroxide procedure was used for non-engorged ticks. The present work aimed to evaluate the inactivation activity and related immune potential of the Cobra venom-derived LAO enzyme compared to the currently used inactivants (BPL and formalin) for both animal and human vaccines. The RVF virus was completely inactivated within 6 hrs, 4 hrs and 2 hrs after treatment with Formalin, LAO and BPL, respectively. An efficient and rapid DNA minipreparation modified method for frozen samples was developed for five tropical tree species: Copaifera langsdorffii, Hymenaea courbaril, Eugenia uniflora, Tabebuia roseo alba and Cariniana estrellensis. This procedure that dispenses the use of liquid nitrogen, phenol and the addition of proteinase K, is an adaptation of the CTAB-based DNA extraction method. The modifications included the use of PVP to eliminate the polyphenols, only one chloroform-isoamyl alcohol step and the addition of RNase immediately after extraction with chloroform. The present study was designated to evaluate the antibacterial activities of aqueous and ethanol:water extracts from leaves, roots and stem bark of Psidium guajava L. The antibacterial activities of the extracts against bacteria were tested by using both microdilution assay. The aqueous extracts of P. guajava leaves, roots and stem bark were active against the gram-positive bacteria Staphylococcus aureus (MICs=500, 125 and 250 µg/ml, respectively) and Bacillus subtilis (MICs=500 µg/ml), and virtually inactive against the gram-negative bacteria Escherichia coli and Pseudomonas aeruginosa (MICs >1000 µg/ml). An experimental evaluation of the effectiveness of beach ashtrays in preventing marine contamination BABT

In this study, hypotheses concerning the use of beach ashtrays were experimentally tested. Results indicated that the mean rate of abandonment of this equipment was low (1.5%). The mean amount of cigarette stubs (3.4 items/ashtray) was greater than mean amounts of other types of litter. People with different socioeconomic profiles had different perceptions regarding the issues associated with beach debris. These results indicated that beach ashtrays could be useful to prevent the contamination of these environments and that differences in socioeconomic characteristics of beachgoers could partially explain the differences in perceptions regarding the presence of waste on the beaches. Buzhong Yi Qi Wan (Buzhong) is a medicinal herb widely used in Traditional Chinese Medicine to treat the digestive and circulatory systems. Red blood cell and plasma proteins labeled with technetium-99m (99mTc) are used in nuclear medicine. The aim of this work was to investigate the effects of an aqueous Buzhong extract on the labeling of blood constituents with 99mTc. Heparinized blood (Wistar rats) was incubated in vitro with different Buzhong extract concentrations and 99mTc-labeling was performed. The complexometric method is usually applied to quantitative calcium determination in different materials; however the application of this method to calcium determination in molluscs shells infers significant interferences to the results. The snail Bradybaena similaris, a terrestrial gastropod, was used as experimental model to the improvement of this method. The shells were calcinated and dissolved in nitric acid, the hydrogen peroxide was also used to clarify the medium after the acid addition. An inexpensive alternative equipment for the plant material embedding in the paraffin under the vacuum BABT

The present work describes an equipment constructed using inexpensive material for embedding the plant material in the paraffin under the vacuum, using an oven and a vacuum pump. The equipment was tested using the samples of Rodriguezia venusta (Orchidaceae) buds embedded in paraffin, where half of the samples were submitted to the vacuum by the equipment during the embedding. The material was sectioned with a rotary microtome, obtaining full series of quality sections. The control was hard to section with the microtome, obtaining damaged sections due the air bubbles, making the ribbon formation difficult. These results proved the effectiveness of the equipment, making it a practical, inexpensive and more portable solution for newly established laboratories.
